Days of our Lives is set in the fictitious Midwestern town of Salem. The core families are the Bradys, the Hortons and the DiMeras, and the multi-layered storylines involve elements of romance, adventure, mystery, comedy and drama.
The results of pregnant Nancy's amnio-test impress Craig, who refuses Colin's offer to tell her and Chloe- he tells them and Brady she's in perfect health and an ideal match for Chloe, but afterward asks gynecologist Dr. Bader for details. Rolf ('professor Putnam') advises the twins, who can't concentrate on their genetics studies anyway, to take a break and join Shawn, who is still worried sick about Hope; Bart happily reports their arrival, and indeed they strongly reject any suggestion their DiMera benefactors could be involved in anything bad like Stefano; Shawn practically throws them out, to Bart's horror they run away from the car and deliberate whether to stay at the mansion. Hope worries after seeing Billie talking with Larry, who actually concludes his unreliable police plant is probably best killed, then tells Hope she is officially dead. Cameron uses a restraining order to keep Bo away from Lexie; Roman has to keep him back, then grills Billie about her whereabouts,
Belle is delighted to hear from Nancy and Chloe at the mall she'll get a sister and her hair is growing back. Craig visits Brady to tell him they must find Sykes for Nancy's sake; he agrees and surprisingly reports to have seen him in Salem, but first gets Craig's approval for another plan for Chloe. Victor comes read Hope's letter; Tony tells them Lexie is innocent, Kiriakis takes DiMera apart to propose ceasing their destructive family feud, but they end up exchanging threats. At the pier, Shane Donovan playfully overpowers Billie, secretly watched by Larry who hears them mention ISA, but they hear him make a noise, he slips off; she tells she was to enter the Salem police to test if Bo would make a good ISA-recruit to investigate DiMera, reporting to agent Ross- he tells that's impossible, Ross's corps was found in the Seine two years ago, showing his picture. Abe tells Lexie she must show Hope's body to get 'just' life; after he left, Bo enters her cell to make her talk by force
On the plane with Chloe, Brady plays hot director/producer Bradford Frame, who promises to make her dream come true in Hollywood, as his dream is being with her. Céleste brings 'collapsed' Lexie to hospital, where a male nurse awaits her according to plan. In the pub, the Brady clan keeps worrying, Shawn in utter fury, blabbing out to Salem Gazette reporters the DiMeras killed Hope as revenge for Lexie loosing Zach. Jen tells Jack she'll take a short leave from the paper; Harold is sent to the hospital for the Spectator, and when he interviews Céleste she expresses a premonition disaster is about to strike Lexie. Abe tells Bo the commissioner has already suspended him, but before he and Roman can interrogate Lexie she has disappeared, two men rush her to the roof where a helicopter will pick them up. Larry is back with Hope, who realizes he shot Billie when he says having removed the last obstacle to his revenge for his prison term: Lexie's execution and Hope's lifelong incarceration ...
On her trip with Brandon, Sami is delighted with the suite and his hot body; after overcoming her nagging about Lexie, they make love. On the island, Marlena and John have scuba-dived to the DiMera place, which is well protected but not match for John's burglary training; inside, the estate seems abandoned since years. Marlena nearly falls into a secret hatch, stares into a sea painting and sneaks trough a secret fireplace entrance. At the hospital, Roman lets only Shawn, Belle, Jack and Jen in, not Harold. Craig finds strangler Bo broke Lexie's neck; Abe and Céleste are beyond themselves; Shane and Roman arrest Bo.
Before Tony DiMera is brought in for questioning Roman questions Shawn. Sami tells Brandon she knows about his and Lexie's night together. Hope's condition has become worse and she sees Larry Welch practically everywhere.
John is interfering with the Colin Murphy case. Chloe suspects her leukemia is coming back.
Bradys and Blacks gather at Colin Murphy's wake, but do more badmouthing then mourning, as Belle points out uncomfortably while worrying about Shawn's silence, as he and Hope do about each-other's. When the twins find the DiMera family bible, Tony is relieved it's not Stephano's secret diary and insists the present, which they are, builds on the past, when John ruined several DiMera lives, but insists they should make the best of their own. Hope has another Larry-apparition, the real one watches outside, planning true grief for them; Marlena and Bo try to get her in therapy, she lies even to Shawn about overhearing Colin planning to shoot Bo. Brady can't stop Chloe worrying about her possible cancer-relapse. Shawn Sr rages at Tony DiMera who turns up with the twins (as Cassie couldn't wait to see Shawn), thinking Tony murdered Colin, so the old fool is told about John being his sibling and gets himself worked up to a heart-attack.
Roman and Bo try to convince Caroline that Shawn Sr. will recover, speculate about some link with the old secret in Ireland, and reproach John not to have told them he's Tony's half-brother. Cassie wants Belle's job, supporting Shawn; Tony warns the twins Bradys will always turn on DiMeras, and indeed John does so till Marlena pulls him away. Rex decides they still go to hospital, for Shawn's and Marlena's sake, but warns Cassie not to risk her friendship with Shawn by rivaling Belle. When Bart reports John no longer petitions to have Daphne dug up, Tony believes Marlena has become his unexpected ally and swears to destroy the Brady-clan, for one thing with the forged letter from Colin. Once the tests are conclusive, Dr. Craig Wesley releases Shawn Sr. to the family, stressing medication and no stress. When Marlena says she wants the feud with the DiMeras to end with her generation, John answers it's going to end soon, but that takes a war.
Lexie and Sami run into a difficulty at the hospital. Bo pushes Hope to tell him what's wrong. Chloe gives Shawn and Belle the news about her father. Kate and Roman find they have certain things in common.
Tony tries to use Sami to get to Marlena. Hope tells Bo about her problems. John has trouble accepting that he is related to a DiMera.
Marlena tells Tony she won't be a mother for the twins, who she believes to be ruthlessly abused as rarely ideal lab rats by Stefano and their own mother who probably sold them, nor neglect her own family; she is given a photo of the twins and runs off with it. Even in Jack's arms, Jen can't shake the memory of Colin kissing her, nor Hope the image of his corps; the two couples meet, still equally obsessed; Marlena joins them, hears both stories and accepts to help both women, but can't shake Tony's plea not to abandon the twins as their mother did. Meanwhile Larry enjoys spooking Hope in her garden, wearing corps make-up. Back home, Jen tells Jack she won't sleep alone, nor make love. Roman uses the handcuffs at Tuscany to force Kate into admitting she's attracted to him, she insists he's beneath her; once she's released, Tony sends her champagne, fishing for Stefano's secret, Roman 'coming to her rescue' makes her accept dinner with Tony, but when he resumes runs out on both men.
