Bosom Buddies follows the adventures of Kip Wilson and Henry Desmond, junior ad-men at Livingston Gentry and Mishkin Advertising Agency. Kip was an illustrator, but aspired to be a painter, and Henry, a copywriter and aspiring novelist. When their building is demolished, they dress as women to stay with co-worker Amy Cassidy in her apartment at the Susan B. Anthony Hotel, where men are forbidden anywhere above the lobby. Kip and Henry decide to invent alter egos Buffy and Hildegarde (their fictional sisters) to move into an empty (and cheap) apartment at the hotel; Henry, to write an expose about the experience, and Kip, to get closer to Amy's beautiful roommate, Sonny Lumet, nurse, and part-time dancer.
Henry Desmond and Kip Wilson are imaginative young admen who solve an acute housing problem by donning wigs and dresses to qualify for residence in the Susan B. Anthony hotel for women.
When Kip is discovered in the apartment without his Buffy disguise, Lilly Sinclair, the new manager of the hotel, decides to have the other residents vote on whether Buffy and Hildegarde are evicted or not.
Feuds erupt amongst Amy and Sonny and Kip and Henry.
Henry worries that people don't perceive him to be masculine, so he overcompensates.
The boys try to out-sleaze a rival ad man to land an account. Meanwhile Isabelle leads a self-defense class.
Sonny and a nervous Kip double date with Henry and Ruth's purple haired niece who calls herself Andrea Puss.
After experiencing life as an unattractive woman, Henry realizes that he himself has been shallow in only dating women for their looks, so he asks out Amy.
Amy instigates an elaborate charade with her friends to get back at a snobby congressional aide who dumped her at a costume party.
Amy employs unusual tactics when she pitches an ad for Mountain Pine mouthwash.
Sonny decides to go home to Michigan, so Kip entices her to stay by getting her a starring role in a TV commercial.
Kip and Henry (as Buffy and Hildy) subdue two criminals that are robbing the hotel, drawing the attention of the media, and Henry's mother.
Kip's artwork inspires Henry to have it displayed in a special showing inside a local art gallery as an original birthday present for him. Some attending patrons though , are not impressed with it.
Kip quits his job rather than change a campaign to please a client and winds up selling hot dogs on a street corner.
When Ruth pushes an evening of home entertainment on Kip and Henry, they realize she may not have any friends. Meanwhile Sonny, Amy and Isabelle care for a box full of kittens.
Buffy and Hildy are nearly unmasked, so Henry writes about the incident and keeps trying to improve the story by changing the tone.
The guys take jobs bussing tables to afford Isabelle the opportunity to sing in a night club.
A blind hospital patient who can see right through Kip falls for Sonny. Meanwhile Lily is hospitalized for mono and Buffy and Hildy work at the hospital as candy stripers.
Henry becomes jealous of Kip's revived friendship with his childhood friend, now a rock star.
Kip and Amy agree to help each other get Sonny and Henry.
