Bosom Buddies follows the adventures of Kip Wilson and Henry Desmond, junior ad-men at Livingston Gentry and Mishkin Advertising Agency. Kip was an illustrator, but aspired to be a painter, and Henry, a copywriter and aspiring novelist. When their building is demolished, they dress as women to stay with co-worker Amy Cassidy in her apartment at the Susan B. Anthony Hotel, where men are forbidden anywhere above the lobby. Kip and Henry decide to invent alter egos Buffy and Hildegarde (their fictional sisters) to move into an empty (and cheap) apartment at the hotel; Henry, to write an expose about the experience, and Kip, to get closer to Amy's beautiful roommate, Sonny Lumet, nurse, and part-time dancer.
Kip's ill timed decision to tell Sonny the truth about his alter ego causes an international incident.
Henry impulsively buys a TV commercial production house and later learns it's heavily in debt.
At their class reunion, Henry relives a painful incident with a deaf girl while Kip tries to flaunt Sonny in front of all of his former classmates.
After berating Kip for being irresponsible with money, Henry buys a stolen VCR and is thrown in jail.
Kip and Henry are nearly crushed in an elevator accident and subsequently decide to experience some wild fantasies before they die.
Kip and Henry drop a water balloon on a car that turns out to belong to Richard Nixon. After being questioned by the Secret Service, Kip is so shaken that he gives up practical jokes.
Depressed after breaking up with another girlfriend, Henry goes to a video dating service, where he meets a beautiful Satanist; Kip tries to lure Sonny into bed.
Henry starts to date an interior decorator that nobody else can stand.
To prove their deep love for Sonny and Henry, Kip and Amy go to a tattoo parlor.
Ruth quits her job and comes to work full time at 60 Seconds Street. When feuds erupt between Ruth and the boys, Amy realizes her stake in the company is significant.
Kip and Henry produce an episode length cable variety show, including an appearance by their special guest star and kidnap victim Penny Marshall.
Henry saves the life of a little girl who turns out to be the granddaughter of a mob boss.
A middle aged man falls in love with Hildy, so Henry fakes Hildy's death.
After a spat with his wife, Kip's dad seeks refuge with Kip and Henry at the hotel; Kip must explain his lifestyle to his father.
To cheer up Isabelle, Henry recounts his college days and the story of how the boys began working for Ruth.
Kip and Henry's weekend at a secluded cabin yields hunger and fears of death. Meanwhile Ruth accidentally lets a tiger loose in New York City.
Kip is furious when he thinks Henry has acted improperly with his kid sister.
Kip, Sonny, Henry and Amy imagine what their lives will be like as senior citizens.
